About the position

We currently have a vacancy at the Group of Molecular and Cellular Cardiology (GMCC) for a PhD Fellowship. This is a 3-year temporary position within the field of cardiac physiology and molecular medicine. We seek a talented and enthusiastic person with experience in medicine, physiology and molecular medicine.

The hosting research group GMCC is located at the Department of Circulation and Medical Imaging, Faculty of Medicine & Health Sciences NTNU and St. Olavs University Hospital in Trondheim, which offers a unique research environment for translational medical research. The research unit is integrated in St. Olavs University Hospital, and is now co-located with the cardiology and cardiothoracic surgery units. Both NTNU and St. Olavs Hospital have excellent infrastructures for clinical and experimental research with state-of-the-art equipment and laboratory technicians and engineers with considerable knowledge and time-allocation dedicated to the laboratory methods. Main duties and responsibilities

Our research projects aim to establish novel insight into the cellular and molecular mechanisms underpinning heart failure with especial focus on processes regulated during and after myocardial infarction. For the characterization of cellular and molecular mechanisms, we use material from different sources: human cardiac biopsies obtained during surgery, experimental comparative animal models as well as cardiomyocytes maintained and treated in culture.

The topic of the current PhD project involves ischemic heart disease and the potential for a class of small non-coding RNAs, called microRNAs to modulate the outcome during and after ischemia reperfusion. In the initial part of this project we have identified several specific microRNA altered in patients with post infarction heart failure. Identifying the significance of these regulated microRNAs, and the cellular and molecular mechanisms they target, will be a key part of the project. Potentially, targeting these specific miRs may benefit the development of novel treatment strategies for heart failure.

The work will involve modulation of expression levels of specific microRNA both in experimental animal models as well as in isolated cardiomyocytes in culture. Key parameters will be assessment of structural changes of the heart, cardiac function and electrophysiological properties using both in vivo and ex vivo models of myocardial infarction. In cardiomyocytes in culture the work will involve established models of hypoxia-reoxygenation with assessments of cellular properties of cardiomyocyte function, calcium handling, electrophysiology, mitochondrial respiration, cellular survival and viability in addition to transcriptomic, proteomics and cellular signaling involved in the potential protective effect of microRNA modulation.

Qualification requirements

The PhD-position's main objective is to qualify for work in research positions. The qualification requirement is completion of a master's degree or second degree (equivalent to 120 credits) with a strong academic background or equivalent education with a grade of B or better in terms of NTNU's grading scale. Applicants with no letter grades from previous studies must have an equally good academic foundation. Applicants who are unable to meet these criteria may be considered only if they can document that they are particularly suitable candidates for education leading to a PhD degree.

We are searching for a person with a powerful internal motivation to identify physiological and cellular mechanisms of the heart in both experimental animal and human studies. The successful candidate must have:

• A master's degree or an equivalent education in a relevant field such as public health, health sciences, epidemiology, biostatistics, genetics, molecular biology, biotechnology, systems biology or a medical degree
• Experience in quantitative statistical analysis and academic writing
• A strong academic record with a weighted average grade of at least B from master's or equivalent education, in accordance with NTNU's grading system
• Proficiency in spoken and written English
• The candidate must have extensive experience in general laboratory work and master work with cell cultures in addition to general biochemical and molecular techniques and analyses for transcriptomic and proteomic analyses

The appointment is to be made in accordance with the regulations in force concerning State Employees and Civil Servants and national guidelines for appointment as PhD, postdoctor and research assistant

Personal characteristics

• Previous research experience and/or publication
• Experience with research in cardiac physiology, especially within myocardial infarction, ischemia reperfusion injury and coding/non-coding RNA
• Experience with experimental animal research including physiological measures of cardiac function both in vivo and ex vivo
• Previous experience with RNA sequencing and proteomics analyses as well as bioinformatics analyses
• Personal motivation and proven ability to work both independently and in a team, as well as potential as a researcher
• Excellent communication skills, both orally and written

Salary and conditions

PhD Candidates (code 1017) are remunerated with a gross annual salary of minimum NOK 449 400. Depending on qualifications and academic background the salary can range between NOK 449 400 - 505, 800. In addition to taxes, a further 2% is deducted for the Norwegian Public Service Pension Fund.

The period of employment is 3 years. Appointment to a PhD position requires admission to the PhD programme.
